PURPOSE:To obtain a highly purified burst-formation promoting factor effective to promote the erythropoiesis and expected to be useful as a remedy for anemia, by treating human urine by column chromatography consisting of a combination of ion-exchange chromatography, gel-filtration and reversed phase chromatography. CONSTITUTION:Human urine, especially urine of anemic patient is subjected to the treatment by column chromatography consisting of the combination of ion-exchange chromatography (e.g. DEAE-Sephadex A-50, A-25, etc.), gel- filtration (e.g. Sephadex G-100, Sephacryl S-200, etc.) and reversed phase chromatography (e.g. phenyl-Sephalose) to separate and collect a burst-formation promoting factor having high purity. The urine left after the separation and removal of erythropoietin from human urine can be used as a raw material of the high-purity burst-formation promoting factor, by treating the urine with cold ethanol and subjecting the resultant precipitate to the above column chromatography.
