PURPOSE:To enable efficient separation and analysis, by adding bivalent metal ion able to form a complex with a phosphate group of nucleotide to a mobile phase in the separation and analysis of nucleotide by a liquid chromatography. CONSTITUTION:In the separation and analysis of nucleotide such as adenosine- monophosphate with a high-speed liquid chromatography apparatus, as separation column and mobile phase, a column for anion exchange chromatography and a mobile phase or a column for opposite phase chromatography and a mobile phase are used. Then, when a bivalent metal ion able to form a complex with a phosphate group of necleotide is added to these mobile phases, a complex of the nucleotide is formed in the mobile phases. This can weaken the intensity of a negative load of the nucleotide itself thereby enabling quick and efficient separation and analysis of nucleotide in the anion exchange chromatography.
