PURPOSE:To separate riboflavine-5'-phosphate in high purity, by reacting a crude riboflavine phosphate in the presence of a phosphatase enzyme. CONSTITUTION:A phosphatase of microorganism belonging to Streptomyces genus (an actinomycete) or belonging to Penicillium genus (a fungus), and having high capability to decompose a riboflavine phosphate other than riboflavine-5'- phosphate, is prepared beforehand. A crude riboflavine phosphate containing riboflavine-5'-phosphate and other riboflavine phosphates is incubated in the presence of the above phosphatase enzyme to effect the selective hydrolysis of the riboflavine phosphate other than riboflavine-5'-phosphate, and the objec tive riboflavine-5'-phosphate is separated from the reaction mixture by conven tional separation and purification means. The phosphate compound is important as a coenzyme for various enzymatic reactions in the living body.
