MONOCLONIC ANTI-ALPHA-AMYLASE ANTIBODY OF ALPHA-AMYLASE USING THE SAME

摘要

PURPOSE:To obtain a monoclonic anti-alpha-amylase antibody having high specificity to S-amylase and to obtain a method for the highly accurate fractional quantita tive analysis of alpha-amylase using said antibody, by using a combination of two kinds of monoclonic antibodies. CONSTITUTION:It is considered that a cell producing an antibody capable of discriminating the extremely slight difference from the aspect of the protein structures of S-amylase and P-amylase is present in a large number of antibody- producing cells obtained by immunizing S-amylase or P-amylase and the objective cell is selected and cloned as a hybridoma to obtain a monoclonic antibody having high specificity to S-amylase. A drawing shows fluorescent intensity generated when polystyrene beads are treated with a monoclonic anti-alpha-amylase antibody solution and white dots show S-amylase and black dots show P-amylase and this measuring method can be said to be perfectly specific to S-amylase.
