PURPOSE:To obtain insulin and elastase useful for pharmaceutical purpose, by separating the above compounds at the same time from swine pancreas. CONSTITUTION:A swine pancreas (e.g. fresh swine pancreas, defatted pancreas, frozen pancreas, etc.) is preferably homogenized e.g. with a homomixer. The homogenized pancreas is added to a 50-80V/V% aqueous solution of a water-soluble organic solvent (e.g. acetone, ethanol, etc.) and treated at 8.0-9.5pH and -5-+5 deg.C. Insulin in the swine pancreas is extracted to the aqueous solution of the water-soluble organic solvent under the above pH condition, and elastase is separated in the form of precipitate. The insulin can be separated by adjusting the supernatant liquid of the above aqueous solution to 4.0pH with sulfuric acid, evaporating acetone in vacuum, and concentrating the solution by ultrafiltration. EFFECT:Both compounds can be produced at the same time in high yield.
