PURPOSE:To purify the titled lyophotoxin, by concentrating a culture supernatant containing lymphotoxin obtained by cultivating a cultivated strain cell derived from human lymphatic system in a serum-free medium, purifying it by gel filtration, and concentrating it further. CONSTITUTION:A cultivated strain cell derived from human lymphatic system such as cultivated strain cell ALIE derived from human malignant lymphoma is used as a LT production source, and the cultivated strain cell is cultivated in a serm-free medium for about 3 days. The prepared supernatant liquid is concentrated by ultrafiltration 5-100 times, and purified by gel filtration (Sephadex or Sephacryl is used). The purified LT is further concentrated by ultrafiltration 5-100 times, to give human LT (lymphotoxin). It is required to select the ratio of concentration before gel filtration suitable for gel filtration treatment and the ratio of concentration after the gel filtration to make antitumor effect maximum. |
