摘要 |
PURPOSE:To make it possible to produce urokinase using the recombinant technique, by cultivating an animal cell transformed with a plasmid containing a specific DNA sequence prepared from an established cell derived from human renal cell. CONSTITUTION:mRNA is obtained from a human renal cell, and a DNA sequence expressed by the formula (only part is shown) is obtained using the above-mentioned mRNA as a template. An animal cell transformed by a plasmid containing the DNA sequence incorporated therein is cultivated to give a single- stranded prourokinase having added saccharide chain thereto. The above- mentioned urokinase is constituted of for example signal peptide consisting of 20 amino acids followed by 411 amino acids. >=80 bases or more 5'-noncoding regions and >=800 bases or more 3'-noncoding regions are present in the urokinase mRNA. |
