PREPARATION OF SURFACE ANTIGEN OF HEPATITIS B VIRUS

PURPOSE:To produce the titled antigen useful also as a raw material of vaccine, by culturing a cell strain huGK-14 originated from human hepatocarcinoma and capable of secreting a large amount of hepatitis virus B surface antigen directly into a culture medium and proliferative even in a serumfree medium, and separating the antigen from the culture liquid. CONSTITUTION:Subculture of human hepatocarcinoma cell strain huSP is repeated in a mutagenic medium and a serumfree medium, and a strain capable of producing a large amount of hepatitis virus B surface antigen (abbreviated as HBsAg) is cloned by detecting the production of HBsAg in the medium. The cell strain huGK-14 obtained by the cloning is cultured. The mutant cell strain huGK-14 secretes a large amount of HBsAg directly in the medium, is proliferative even in a serumfree medium, and can be grown well in the Williams E medium. USE:HBsAg useful as a raw material of vaccine and a diagnostic agent of the infection with hapatitic virus B can be produced in large quantities.
