PURPOSE:To prepare an immobilized enzyme having high immobilization ratio and enzymatic activity development ratio, by bonding an enzyme to a carboxyl- containing polymeric compound through ionic bond, forming a covalent bond therebetween, and breading the ionic bond. CONSTITUTION:1-10g of a carboxyl-containing polymeric compound is immersed in 100ml of an aqueous solution of enzyme containing 100-5,000 units of enzyme per 1ml, and the mixture is stirred ar shaken at 5-40 deg.C for 1-24hr to bond the enzyme to the compound through ionic bond. 20-40ml of an acetate buffer solution containing 0.1-2M a water-xoluble carbodiimide is added to the reaction product and stirred at 15-35 deg.C for 5-24hr to form a covalent bond. Finally, the ionic bond is broken with a 1-3M acetate buffer solution of 4.1-5.0pH, and the product is washed with water.
