PURPOSE:To enable the effective recovery of heteroprotein without being left in the cell of bacteria such as E.coli, by secreting a heteroprotein in a periplasm using Gram-negative bacteria. CONSTITUTION:A heteroprotein is secreted in a periplasm by using a system originated from Gram-positive bacteria as an expression system gene, and Gram- negative bacteria as the host. In the secretion to the periplasm, the signal peptide is incised and a mature protein free from methionine at the N-terminal can be produced. Human interferon-alpha2 can be produced as the heteroprotein by using an alpha-amilase promoter originated from Bacillus natto as the expression- system gene of Gram-positive bacteria and E.coli as the host. |
