PURPOSE:To determine Mg in a specimen in high selectivity and detection sensitivity, by reacting the specimen with two kinds of enzymes containing hexonase, and comparing the absorbance of produced nicotinamide adenine dinucleotide phosphate hydride at a specific wave length with the absorbance of Mg having known concentration. CONSTITUTION:A specimen such as untreated serum is made to react with a hexonase and glucose-6-phosphate dehydrogenase (G6PDH) to produce nicotinamide adenine dinucleotide phosphate hydride (NADPH). the absorbance of the NADPH is measured at a wavelength of 340nm, and compared with the absorbance of Mg having known concentration to determine the Mg concentration in the specimen. The determination of Mg concentration can be carried out accurately by this process. The process has high selectivity compared with conventional xylidyl blue process, and the scattering of the obtained values is small.
