Whole cells of E. coli KFCC-84-3 high in penicillin G acylase were immobilized in carrageenan, cellulose acetate, agar, or other insoluble polysaccharides. Their ability to produce 6- aminopenicillanic acid from penicillin G in alkaline pH was investigated. Thus, E. coli cells grown on the medium contg. trytone 1, yeast ext. 1, fructose 1, beef ext. 0.5, NaCl 0.25% at 30≦̸C for 12h. were harvested, centrifuged, re-suspended in 100ml of 0.1M phosphate beffer, mixed with a soln. of 4g carrageenan in phosphate buffer 100ml and 0.2v/v % poly-ethyleneamine, dried, and crushed to give immobilized prepns. An average 6-aminopenicillanic acid conversion rate of 90% was obtd. in a column with immobilized cells when recycling the reaction mixst. for 2h. at the rate of 10 - 1501/ h.
