PURPOSE:To determine accurately a basic arrangement of nucleic acid, by detecting and correcting the strain, when strain is observed in an electrophoretic pattern of a nucleic acid piece. CONSTITUTION:A base constituting the nucleic acid is divisible normally into 4 kinds adenine (A) as purine; guanine (G); thymine (T) as pyrimidine; cytosine (C). These 4 bases constitute each lane of A, T, G, C by a phoretic separation gel. Now, when it is considered that a phoretic band 4 of the nucleic acid piece of the same molecular weight and higher lane temperature of T, and G and lower A, C lane temperature, the phoretic bands 4 of T, G arrive earlier to a detector 8 than that of A, C. As the arrival time of the phoretic band 4 is roughly proportional to gel temperature, an arrival time difference can be known by measurements of temperatures, of each lane with a thermo-couple 9. An accurate order of the phoretic bands is determined by introducing a signal related to the arrival time of the band 4 detected by the detector 8 and each lane temperature signal into a computer 15 and correcting the arrival time. |
