6-O-METHYLERYTHROMYCIN A DERIVATIVE

NEW MATERIAL:The oxime compound of formula I. USE:An antibacterial agent. It can be converted to 6-O-etylerythromycin a in high yield. PREPARATION:The objective compound can be prepared e.g. by (1) reacting the compound of formula II [R<1> is (substituted) benzyl; R<2> is benzyloxycarbony] with usually 1.2-2.4mol-equivalent of methylating agent (e.g. methyl iodide) in an aprotic polar solvent such as DMF in the presence of a base (e.g. sodium hydride) at 0 deg.C - room temperature, (2) reducing the produced 6-O-methyl derivative in an alcohol solvent in the presence of Pd-C in hydrogen atmosphere under agitation to remove R<1> and R<2>, and (3) carrying out the N-methylation of the resultant 6-O-methyl-N-demethylerythromycin A 9-oxime in an inert solvent in the presence of formic acid and excess formaldehyde under heating and stirring.
