<p>The 2-(N-benzyl N-methylamino)ethyl ester hydrochloride of 2,6-dimethyl 4-(3-nitrophenyl) 3-methoxycarbonyl 1,4-dihydro pyridine 5-carboxylic acid, of formula (I) is made without isolating intermediates by (a) reacting 3-nitro benzaldehyde with methyl acetate in molar ratio 2-2.2 and with NH3 inmolar ratio 1-1.3, in dioxane at b.pt. for 5-10 hours; (b) adding a drying agent, e.g. MgSO4, and activated carbon; (c) adding N,benzyl N-methyl 2-amino ethanol in molar ratio 0.8-1 to the aldehyde, and a basic catalyst, e.g. NaH, to obtain the base of (I); (d) treating with HCl; and (e) recrystallising (I) from aq. alcoholic solvent.</p> |
