PURPOSE:A microorganism in Cryptococcus is cultured and a novel aldose reductase which has ability to reduce ketoses and can utilize NADPH and NADH as a coenzyme. CONSTITUTION:Cryptococcus lactacivorus sp. H131 (FERM 7840) is cultured in a medium containing apropriate amounts of yeast essence and xylose, or cultured first with a medium containing yeast essence and glucose, then cultured again with medium containing xylose and ribose. A uniform aldose reductase A is obtained by ammonium sulfate fraction, ion-exchange chromatography and other techniques. The resultant aldose reductase has ability to reduce ketoses and can utilize NADH or NADPH. |
