PURPOSE:To enhance the analytical sensitivity of amino acid, by removing background beam due to Raman scattering by a fluorescence measuring method using laser beam as a light source. CONSTITUTION:The amino acid-containing specimen injected from an injection port 24 is sent to a specimen trap column 1 by a pump 10. Next, six-way valves 21, 22 are changed over and 7-fluoro-4-nitrobenzo-2-oxa-3-diazol solution is sent in said column 1 by a pump 11 while the specimen is sent to a modified amino acid trap column 2 under reaction to trap modified amino acid. Next, the valves 22, 23 are changed over and an eluate constituted of citric acid buffer solution-acetonitrile-tetrahydrofran is sent into a separation column 3 according to a gradient elution method by a pump 12 to separate NBD-amino acid. The separated NBD-amino acid is detected by a fluorescence detector 20 using laser as a light source. |
