PURPOSE:To enable the three-dimensional culture of a plant tissue in a liquid medium in high efficiency, by placing pieces of plant tissue in a culture vessel furnished with a means for supplying a liquid medium and a means for discharging the liquid, and alternately carrying out the supply and discharge of the liquid medium. CONSTITUTION:The cut pieces of plant tissue placed on the bottom of the culture vessel 1 are immersed intermittently in a liquid medium supplied to and discharged from the vessel 1 through the port 2. The liquid medium is supplied to or discharged from the vessel by the pump 4 attached to the line 3 connecting the culture vessel 1 and the liquid medium reservior 5. The medium may be aerated through the aeration holes 6 or agitated by an agitation means.
