Sepn. of serotonin (I) from coffee wax comprises subjecting the wax in soln. to aq. alkaline hydrolysis with a strong base under an inert atmosphere. Then the (I) is isolated. As solvent for the coffee wax, an ether of formula R-(OC2H4)x-O-CnH2n-OH (II) is used. In (II) R = H or 1-4C alkyl; x = 0 or 1; n = 2,3 or 4. Provided that x is not 1 when n = 3 or 4. - (2) Prepn. of N-acetylserotonin (III) comprises acetylation of (I) to give N, O-diacetylserotonin (IV), which is selectively hydrolysed with alkali in aq. lower alkanol to hydrolyse the O-acetyl gp. (3) Prepn. of mexamine (V) comprises deacetylation of melatonin (VI) in hot alkaline soln. contg. a water-insoluble alcohol. The alcoholic phase is washed with water and then acidified with HCl.
