PURPOSE:To prepare a liposome containing insulin in high concentration in aqueous phase of liposome and having excellent stability with time, by adding a specific sugar fatty acid ester to the membrane component of liposome composed of a phospholipid and cholesterol. CONSTITUTION:An insulin-containing liposome is prepared by adding the sugar fatty acid ester of formula I [r is OH or group of formula II (R<1> is 11-19C alkyl); at least one of the group R is group of formula II] (an ester commercially available from Dai-ichi Industrial Chemical Co. Ltd. in the name of F-10, F-20, F-30, etc.) to the membrane component of liposome composed of a phospholipid (e.g. phosphatidylcholine, phosphatidylethanolamine, phosphatidylserine, etc.) and cholesterol. The amount of the ester is 10-100wt% based on the phospholipid. The molar ratio of the phospholipid to cholesterol constituting the membrane component is preferably 7:(2-7). A sugar fatty acid ester having an HLB of 6-15 gives high insulin-incorporation rate. |
