PURPOSE:To produce a creatine amidinohydrolase having wide optimum pH range, excellent thermal stability and small Km value, by the microbial technique using a microbial strain belonging to Actinobacillus genus. CONSTITUTION:A microbial strain belonging to Actinobacillus genus and capable of producing creatine amidinohydrolase, e.g. Actinobacillus CRH-1271 (FERM-P No.7721), is used in the present process. The strain is cultured in a nutrient medium [preferably containing creatinine (derivative) or creatine (derivative)] at about 25-35 deg.C and about 6.5-7.5pH aerobically for about 1-2 days. The microbial cells separated from the culture product are disintegrated to obtain crude enzyme liquid, which is treated by conventional method (salting-out, precipitation with organic solvent, dialysis, gel-filtration, etc.) to produce the objective creatine amidinohydrolase. |
