<p>A process is claimed for determining glutamate oxalacetate transaminase (GOT) or glutamate pyruvate transaminase (GPT) in biological fluids which contain endogenous substances capable of being oxidised in the presence of glutamate dehydrogenase (GDH) with simultaneous formation of NADH. The process comprises (a) incubating the test fluid with NAD and GDH to oxidise the endogenous substances; (b) incubating the mixt. with pyruvate and lactate dehydrogenase to oxidise NADH to NAD and reduce pyruvate to lactate; (c) inactivating the LDH; (d) incubating the mixt. with alpha-ketoglutarate, NAD, GDH and the appropriate transaminase substrate (L-aspartate or L-alanine); and (e) measuring the amt. of NADH formed. The process suppresses interence from endogenous amino acids such as glutamic acid, leucine, isoleucine, valine, methionine and alpha-aminobutyric acid.</p> |
