PURPOSE:To assay a recombiant human interferon beta(gamma IFN-beta) simply and at a high accuracy, by an enzyme-immunoassay employing a solid phase double sandwich method. CONSTITUTION:First, when an antigen (gamma IFN-beta) is made to react with an antibody B obtained by making a polyclonal antibody thereagainst insoluble, an antigen-antibody composite (c) is formed. Then, after separated from the reaction solution, the composite (c) undergoes reaction with a fixed amount of monoclonal antibody D against an antigen A to form an antigen-antibody composite E. Moreover, the composite E is separated from the reaction solution and allowed to react with an enzyme labelled antibody F as such against immune globulin, making the enzyme label E exist isolated beyond the bonding capacity. Then, the enzyme labelled antibody F bonded to the former composite E is separated from those F isolated to measure the activity of the enzyme made insoluble. At the same time, the reference substance with the known concentration in used to prepare a reference curve and the amount of an unknown antigen is determined by use thereof. |
