PURPOSE:To enable the secretion and production of an oligopeptide from a bacterial cell, using a vector prepared by introducing a DNA coding the objective oligopeptide to a plasmid containing a leader gene and an alpha-amylase structural gene. CONSTITUTION:A DNA coding the objective oligopeptide (e.g. angiotensin I, bradykinin, insulin, etc.) is linked to an alpha-amylase gene containing a leader gene and an alpha-amylase structural gene, to effect the secretion of the fused protein consisting of alpha-amylase and the oligopeptide from the bacterial cell. In the conventional process, the produced oligopeptide is retained in the bacterial cell and accordingly it is necessary to disintegrate the bacterial cell resulting in the difficult purification. In contrast to the above, the oligopeptide is secreted from the bacterial cell in the present process, and the separation and purification of the objective compound can be carried out easily. |
