<p>A cellular organism useful in an assay for determining the metabolism of a compound, the organism comprising in the genome of its cell or at least one of its cells a coding sequence for expressing a polypeptide having the function of a naturally-occurring protein which is involved in the alteration of the metabolism, mutagenicity or toxicity of a compound under the regulatory control of a suitable promoter, the combination of the coding sequence and the promoter not normally being found in the said cell of the said organism. Preferably, the protein is a P450 cytochrome-dependent enzyme. The organism may be yeast (in which case a mammalian NADPH:cytochrome P450 reductase or a hybrid yeast/mammalian P450 reductase can usefully be encoded as well), a rodent (in which case expression in the skin using a keratin promoter is preferred, optionally with co-expression of a glutathione S-transferase) or a Drosophila fly.</p> |
