NEW MATERIAL:Antitumor substances FNF I, II OR III (their physicochemical and biochemical properties are given in table I ; molecular weight 10,000; glycoprotein). USE:Antitumor substance. PREPARATION:Human fibroblasts are transformed into tumor cells (such as SV40 transformed cell) and the cells are made in a confluent state. The supernatant of the cell culture mixture is combined with cold saturated ammonium sulfate solution and, after left standing subjected to centrifugation under cooling. The precipitate is dissolved in a phosphate buffer. NaCl solution and subjected to column chromatography with Sephadex G100 to effect elution and fractionation with PBS. Each fraction is evaluated on the inclusion active of [<3>H] thymidine into 3T3 fibroblasts. Active fractions are collected, freeze-dried to collect the antitumor substance. The substance has been named FNF (fibroblast- originating necrotic factor) and acts only on tumor cells to inhibit them. |
