摘要 <p>PURPOSE:To provide a DNA sequence coding an amino acid sequence equivalent to urokinase derived from human urine, by using an mRNA obtained from the human kidney cell as template, inserting a vector to the cDNA prepared by using the template, and inserting a plasmid to a host. CONSTITUTION:The urokinase mRNA separated and purified from the human kidney cell is treated with SI nuclease to remove the single stranded fraction. The obtained double stranded fraction is subjected to the sucrose density gradient centrifugation to collect the dcDNA fraction having high molecular weight. About 1/5 of the double stranded cDNA is added with the tail of poly(C). Separately, the tail of poly(G) is added to the plasmid pBR322 of Escherichia coli. Both products are linked together by annealing. The pBR322 plasmid inserted with dscDNA and prepared by the above process is introduced into Escherichia coli to obtain a transformation.</p>
