Dipeptide, e.g. Asp-Phe, was obtained by use of the recombinant DNA technology and synthetic genes which are able to express the dipeptide repeatedly. Thus 3 kinds of dodecanucleotide, 5'TTCGACTTTGAT3' (I), 3'AAACTAAAGCTG5' (II), and 3'GAAACTAAAGCT5' (III), which had the Phe Lodon (TTC,TTT) and Asp London(GAT, GAC) were prepd. by post-triesterification. Synthetic genes were prepd. by mixing an equal quantity of (I)) and (II) or (I) and (III) with oligonucleotides, having the restriction enzyme recognition site and ligasing by T4 DNA ligase in the presence of 20mM Tris HCl(pH7.6) contg 10mM dithiothreitol and 0.5mM ATP. Phe-Asp is used as the precursor of aspartam.
