PURPOSE:A monoclonal antibody that has specific reactivity with trophozoite membrane antigen and cyst membrane antigen in Pneumocystis carinii, thus enabling effectively early diagnosis on Pneumocystis carinii pneumonia with high safety. CONSTITUTION:An antigen originating from Pneumocystis carinii, preferably cysts which have been collected from the lungs of mammarian animals infected with Pneumocystis carinii according to the cyst purification method or the supernatant after separation of cyst membrane components is used to immunize mammarian animals such as mice or rats. Then, immune cells, preferably spleen cells are taken out of the animals and fused with myeloma cells of a mammarian animal. The fused cells are cultured in a customary selective medium such as HAT medium to isolate the desired hybridoma. Then, the hybridoma is subjected to limiting dilution to effect detection of the objective antibody and the antibody is monocloned to give the objective monoclonal antibody which specifically reacts with trophozoite membrane antigen and cyst membrane antigen of Pneumocystis carinii.
