PURPOSE:To obtain a tyrosyl oligopeptide in high yield with high reproducibility and carry out easily the subsequent radioactive iodination process, by reacting tyrosine with an oligopeptide at a selected charging molar ratio. CONSTITUTION:(C) An oligopeptide, e.g. synthetic human C-peptide, is reacted with (D) tyrosine in a molar amount within 0.01-10mol range based on one mol component (C) in the presence of (A) tyrosyl-tRNA synthetase (an enzyme catalyzing the reaction; tyrosine+ATP+tRNA tyrosyl-tRNA+AMP+pyrophosphoric acid, optimally obtained from thermostable bacteria, e.g. Bacillus thermophilus) and (B) adenosine triphosphate, preferably at 1:1-1:100 molar ratio between the components (A) and (B) to give a tyrosyl oligopeptide, which is then iodinated with radioactive iodine (<125>I) to afford the aimed substance.
