<p>The process consists of (a) homogenising sheep or pig brain in 70:30 methanol/water with anionic, cationic or amphoteric surfactants at concns. below and above their inorganic critical micellar concns.; (b) purifying the extract by HPLC/TLC or a silica gel column with elution using chloroform/methanol/H2O-0.05 TFA in ratio 60:35:8 as Solvent A, and in ratio 65:25:4 as Solvent B; and (c) de-salinating the ganglioside extract free from contaminants in a C-18 semi-preparative column or C-18 Sep-Pack cartridge, eluting with water and 2:1 chloroform/methanol mixt. - The surfactants can be alkyl sulphate or alkyl ether anionic types, N-alkyl trimethyl ammonium halide or dimethyl alkyl benzyl ammonium halide cationic types, alkyl phenol or ethylene oxide condensates as non-ionic types, or alkyl amido-betaines as amphoteric types.</p> |
