PURPOSE:To enable exact measurement of the number of methane bacteria or the methane forming activity by irradiating excitation light of a specific wavelength region to a specimen having the methane bacteria, measuring the intensity of the fluorescence emitted by the specimen and correcting the same by the turbidity of the specimen. CONSTITUTION:A sample is drawn from a methane fermentation tank 7 by a sampling pipe 8 and is diluted and adjusted by a sample adjuster 12. The adjusted sample is put into a vessel of a sample chamber 13 to prepare the specimen. The excitation light of a specific wavelength region where the material specific to the methane bacteria is made to emit fluorescence is selected by a spectroscope 16 and the excitation light having the wavelength ranging 220-300nm or 380-440nm is used. The fluorescence generated from the specimen is detected by a photoelectron multiplier 21 and the transmitted light is detected by a photoelectron multiplier 24. The photocurrent showing the intensity of the fluarescence and the turbidity is measured by a detector 22 and calculated by a microprocessor 26 and the result thereof is disployed to an output terminal 27. |
