PURPOSE:To recover easily the optical resolving ability of a column having a separation ability deteriorated by continuous use, by washing a separation agent consisting of powdery cellulose with a strong acid, and resolving optically an amino acid by liquid chromatography. CONSTITUTION:Powdery cellulose having the structure of natural cellulose, preferably microcrystalline cellulose is used as a separation agent, and washed with a strong acid, e.g. hydrochloric, sulfuric acid or nitric acid, to carry out optical resolution of an amino acid. The optical resolution is carried out by injecting a mixture of optical isomers of amino acids to be resolved into a chromatographic column, and developing the adsorbed isomers with an eluent, e.g. pyridine or ethanol.
