PURPOSE:To prepare purified titled factor useful as an antitumor agent, by immobilizing one or more kinds of monoclonal antibodies prepared against human tumor necrosis factor in an affinity column, and passing a roughly refined liquid of said factor through the column. CONSTITUTION:A roughly refined liquid of human tumor necrosis factor (e.g. peripheral quadrimonocyte obtained by the cultivation in the presence of a substance having differentiation inducing capability, or the supernatant liquid of the cultivation medium of a cell strain established from the leukemia cell of a certain kind of myelogenous leukemia patient, etc.) is passed through an affinity column containing at least one kind of immobilized and supported monoclonal antibody produced against said factor (e.g. the antibody prepared by culturing a hybrid cell prepared by the cell fusion of a mouse lymphocyte B cell and a myeloma cell). The resultant purified human tumor necrosis factor is a protein having the following characteristics. Amino acid composition, shown in Table I ; molecular weight, 40,000-4,000 (by gel-filtration); isoelectric point, 6.0+ or -0.5; protein. EFFECT:It has excellent carcinostatic activity and low species specificity.
