PURPOSE:To separate CSF effective to promote the differentiation and proliferation of myeloma cell of mammals, in high yield and purity from human urine. CONSTITUTION:Human urine is made to contact with a silicon-containing adsorbent [e.g. SiO2(H2O)n, etc.], and the adsorbed valuable substance is eluted with an alkaline solution of >=9pH. The eluate is made to contact with a cation exchange material under neutral condition to effect the adsorption and removal of impurities. The treated elutant is made to contact with an anion exchange material at 3.5-6pH to adsorb the valuable component to the ion exchange material, and the adsorbed component is eluted with 0.1-0.3M inorganic salt solution. The eluation process is repeated at 6-8pH. The elutant is adjusted to an inorganic salt concentration of 0.6-1.5M, made to contact with a hydrophobic affinity adsorbent at 6-8pH to effect the adsorption of the valuable component to said adsorbent, and eluted with a 0.4-0.15M inorganic salt solution. The elutant is brought into contact with a gel-filtration agent, and the fraction having a relative eluate volume of 1.1-1.7 to obtain high purity CSF.
