<p>Prodn. of a bacterial clone which produces human alpha-1-antitrypsin (AT) comprises first extracting and purifying total mRNA from human liver, then prepg. a fraction enriched in the mRNA for AT. This fraction is used for in vitro synthesis of double-strand cDNA and the DNA fraction enriched in material coding for AT. - Cohesive ends are attached to the DNA then this converted, in vitro, to hybrids with a plasmid vector having complementary ends. The hybrids are used to transform E.coli cells and the transformants contg. human DNA fragments selected. The selected clones contg. AT-DNA are isolated and those which can express AT are identified. Also claimed are double-stranded cDNA coding for AT and the E.coli clones contg. AT-DNA. - AT is useful for treatment of hereditary conditions which make the subject liable to pollutant-induced lung damage (esp. emphysema), partic. when caused by excessive release of proteolytic enzymes. Large amt. of AT can be made by cultivating the transformed bacteria.</p> |
