An enzyme column constitutes a part of a throughflow system comprising a vessel contg. substratum soln., a peristaltic pump and a two-way valve. The valve permits substratum flow to be directed either directly to the flow cell or via the enzyme column. Flow speed is maintained at 0.2 ml/min. The wave conductor in the convertor is of the SiO2-TiO2 type, and the surface relief grid has a space frequency of 1200 lines/mm. The convertor is mounted on a rotating precision location with accuracy of 1 arc minute. - A beam from a He-Ne-laser is directed towards the grid, and to determine the precise angle of approach from the beam, the max. intensity of the transmitted mode (transverse electric or transverse magnetic) is investigated. This is achieved by detecting the light diffused by the transmitted mode with a fibre sond and a photo multiplicator tube, since the diffused light is proportional to that of the transmitted mode. The diffused light is monitored using a lock-in detection process. |
