PURPOSE:To obtain a peptide containing a free carboxyl end in high yield without using a protecting group, by reacting an amino acid with an ester in the presence of a specific enzyme to give a reaction solution, adjusting the pH of the reaction solution. CONSTITUTION:An amino acid (e.g., tyrosine) and an amino acid ester or peptide ester (e.g., L-phenylalanine ethyl ester) are added to water a buffer solution such as tris hydrochloride, etc. containing 0.01-500mM divalent cation such as Mg, Mn, etc. (e.g., MgCl3), 0.001-100mM sulfhydrylating agent (e.g., mercaptoethanol, etc.) or 0.001-100 units/ml pyrophosphatase, and (deoxy) adenosine triphosphate, adjusted to <7pH, aminoacyl-tRNA synthetase obtained from heat-resistant bacterium such as Bacillus stearothermophilus, etc. is added to the reaction mixture, and a reaction is carried out at 30 deg.C to give a reaction solution at <7pH, which is adjusted to >=7pH with NaOH, etc., to obtain peptide.
