PURPOSE:To create a vector expressing calf prochymosin gene which is a heterologous gene in Bacillus subtilis, by utilizing a promotor of an erythromycin-resistant (Emr) gene. CONSTITUTION:A prochymosin gene isolated from pCR702, together with a reading frame, is inserted into a Bucillus subtilis system containing an erythromycin- resistant (Emr) promoter to form an expressed plasmid pKSR101, which is then transformed and introduced into a host of the Bacillus subtilis to afford a transformant strain. A chemical-resistant strain is selected from the resultant transformant strain and cultivated in a nutrient culture medium to recover the prochymosin. |
