发明名称 NOVEL PLASMID ORIGINATING FROM ANAEROBIC THERMOPHILE
摘要 NEW MATERIAL:A plasmid (pSH62) originating from an anaerobic thermophile that is characterized by its molecular weight of about 3.3 megadalton and restriction enzyme cleavage map as shown. USE:It is used as a vector in DNA recombination using an anaerobic thermophile as a host and the use of the resultant vector enables the breeding of an anaerobic thermophile which can produce useful chemicals with high biomass conversion efficiency. PREPARATION:A soil from Ibaragi Prefecture (Japan) is added to YPG medium and cultured under anaerobic conditions containing carbon dioxide at 70 deg.C for about 18hr and further in a YPG roll tube containing 50mug/ml of neomycin. Then, thermoaerbacter No. 62 (FERM P-7493) is obtained from one of the colonies which have grown in the above roll tube. The objective plasmid is collected by lysis of the cells by treatment with lysozyme or SDS.
