PURPOSE:To measure exactly the concn. of the digoxin in the fluid of a living body by conjugating the hapten of a specific digoxin deriv. and a fine particulate carrier insoluble in water and using the same as a reagent. CONSTITUTION:The hapten expressed by the formula I , for example, digoxin-16'- beta-D-glucuronic acid, is conjugated with a fine particulate carrier insoluble in water by amide conjugation. For example, hapten A is mixed with a carboxy latex suspension together with a binder such as N-hydroxysuccinimide, 1-ethyl- 3-(3-dimethylaminopropyl)carbondiimide hydochloride, etc. by which the amide conjugate of the hapten A and latex is obtd. Such conjugate and anti-digoxin serum are mixed and the mixture composed thereof is used as a reagent for quantitative determination of digoxin. The digoxin in the serum and blood plasma is thus quantitatively determined with good sensitivity by the reagent consisting of the conjugate of the hapten expressed by the formula I and the fine particulate carrier insoluble in water, by which the adequate treatment of a heart patient is made possible. |
