NEW MATERIAL:A compound having the following properties; Molecular weight; About 135,000. Isoelectric point; 5.5-5.8 (measured by the two-dimensional electrophoretic method). Content of neutral saccharide; 1.3-1.5% (measured by the phenol-sulfuric acid method). Properties; White odorless powder. Solubility; Soluble in water, 0.01M phosphoric acid buffer solution-saline solution and insoluble in methanol, ether and chloroform. Color reactions; Positive to the biuret, Folin, and xanthoprotein reactions. Ultraviolet absorption; E280nm<1cm>6.47 (1%, water). Disc electrophoretic pattern; Single band having a sharp tip. Subunit constitution; Three subunits having 78,000, 34,500 and 24,800mol.wt. [measured by the sodium dodecyl sulfate (SDS) electrophoretic method]. USE:A clinical test reagent for cancer diagnosis. PREPARATION:Human blood serum is diluted, and 4M ammonium sulfate is added to fractionate the blood serum therewith. The fraction is then dialyzed, adsorbed on a basic anion exchange resin, eluted and subjected to the gel filtration and ion exchange column chromatography to afford the aimed novel protein.
