PURPOSE:To provide a novel vector containing specific three kinds of DNA fragments, giving a large number of copies, keeping excellent stability in a host, and distributed surely from the mother cell to the daughter cell in the cell division of the host cell in subculture. CONSTITUTION:The objective vector CPSMY-4 is proliferative autonomously in the cell of Escherichia coli and contains (A) a DNA fragment having temperature-sensitive mutation to the CI gene of lambda-phage and containing the lambdaNOP-region DNA controlling the copying function, (B) a DNA fragment containing mini-F controlling the proliferation-controlling and distribution system and originated from F-factor plasmid, and (C) a DNA fragment containing the gene controlling the autonomous proliferativity and originated from plasmid PBR322. It has one site incisable with restriction enzyme BalI in the DNA fragment originated from plasmid PBR322. |
