PURPOSE:To obtain lithium salt of a acetyl phosphate of high purity utilizable for enzymic reaction, by reacting acetic anhydride with a phosphate within a specific molar ratio range between the lithium and the phosphoric acid group as a substrate for acetylation reaction in the state of coxisting solid and liquid phases. CONSTITUTION:Acetic acid usually used as a medium is mixed with, e.g. phosphoric acid, an ionic lithium compound, etc. under stirring to prepare a slurry containing a phosphate within 1.5-2.2mol ratio between the lithium and the phosphoric acid group. Acetic anhydride is then dropped thereto and reacted therewith preferably at 30-70 deg.C in the state of coexisting solid and liquid phases to separate the aimed solid lithium salt of monoacetyl phosphate from the riaction mixture.
