PURPOSE:To carry out the determination of amylase in high accuracy, by decomposing maltopentaose with maltose phosphorylase. CONSTITUTION:Maltopentaose is used as the substrate, and is decomposed with amylase. The produced maltose is treated with maltose phosphorylase in the presence of phosphoric acid to produce glucose. Hydrogen peroxide is produced by reacting the produced glucose with mutarotase and glucose oxidase. Amylase can be determined in high accuracy by determining the produced hydrogen peroxide by colorimetry.
