PURPOSE:To obtain a plasmid having pocks-forming ability, and thiostrepton resistance, derived from Streptomyces roseochromogenus of Streptomycetaceae. CONSTITUTION:A thiostrepton-resistant gene range of plasmid of Streptomycetaceae is isolated with restricted enzyme Bel I , it is introduced into a scission point BglII of raw material plasmid. This plasmid has a relative wide host range, and S. roseochromogenus, S.livedans, S. lavendulae, etc. may be cited as the host. This plasmid has thiostrepton resistance, and is useful for cloning a gene from a Streptomycetaceae as a vector by using pocks formation as a selective marker or insertion deactivation of thiostrepton resistance. A vector derived from Streptomycetaceae exists as a relatively large number of copies (about 100 copies) per cell. |
