PURPOSE:To obtain the titled compound in high purity efficiently by easy operation, by reacting a phosphorus acceptor component with a phosphorus donor component in the presence of a hydrophilic solvent, reacting a protecting agent for active sites, and mixing the reaction product with water to precipitate and separate selectively the aimed compound. CONSTITUTION:(A) A phosphorus acceptor component is reacted with (B) a phosphorus donor component to give a reaction product, and active sites, if present therein, are then reacted and protected with (C) a protecting agent to prepare a completely protected nucleotide compound. In the process, the components (A) and (B) are reacted in the presence of a hydrophilic solvent and then with the component (C) to convert the total amount of the component (A) into a protected nucleotide compound. The resultant reaction solution is then mixed with water to precipitate and separate selectively the reaction product, and if necessary the resultant reaction product is dehydrated to afford the aimed compound.
