PURPOSE:To obtain a high sensitivity and high accuracy result, by using a hepatisis B diagnostic kit containing a supported carrier such as a glass bead conjugated with the monoclonal antibody to a hepatitis B core (HBc) antigen and monoclonal antibody-enzyme conjugate to the HBc antigen. CONSTITUTION:For example, 200mul of neutralizing reagent is added to 100mul of a speciment and, after standing at about 37 deg.C for about 1hr, a glass bead conjugated with an anti-HBc monoclonal antibody is added to the resulting solution mixture to perform reaction at about 40 deg.C for about 2hr while the reaction product is washed with a phosphate buffer solution containing 0.1% Tween 20 three times. In the next step, for example, 200mul of an anti-HBc monoclonal antibody- peroxidase conjugate is added to the reaction product to be reacted therewith at about 40 deg.C for about 1hr. Thereafter, the reaction product is washed three times with the phosphate buffer solution containing 0.1% Tween 20. Subsequently, 300mul of an ortho-phenylenediamine substrate solution is added to the washed one to reacted therewith at a room temp. for 30min. At last, reaction is stopped by 1ml of 1N sulfuric acid and absorbancy is measured at 492nm to calculate an inhibition ratio. |
