PURPOSE:To obtain the titled substance in high yield and purity, by treating a purified alpha2-macroglobulin at a specific pH. CONSTITUTION:A purified alpha2-macroglobulin blood serum, e.g. a substance obtained by fractionating a material containing alpha2-macroglobulin such as human blood serum or placenta, with ammonium sulfate, or preferably purifying the fractionated material by affinity chromatography, etc. using anti alpha2-macroglobulin antibody as a ligand, is treated at 2-4ph, preferably 2.5-3pH, more preferably 2.8pH to afford the aimed low-molecular alpha2-macroglobulin (L-alpha2M). The resultant L-alpha2M is an antigenic substance causing IgA renopathy, and the anti L-alpha2M antibody in blood serum of patients with the IgA renopathy can be detected therewith. Thus, the L-alpha2M is useful as serological diagnostic agent for the IgA renopathy. |
