NEW MATERIAL:The bilirubin oxidase having the following physical and chemical properties. Action, oxidizes bilirubin to form a colorless substance via biliverdin and a pale violet substance without producing H2O2; base specificity, active to bilirubin, slightly active to biliverdin and active to phenol, etc.; optimum pH, 5.5; pH stability, stable at 5-9pH and 37 deg.C for 60min; optimum temperature, 50 deg.C; thermal stability, stable at 55 deg.C and 7pH for 10 min; molecular weight, 83,000; isoelectric point, 3.92+0.05; etc. USE:Enzyme for the quantitative determination of bilirubin in serum. PREPARATION:The objective enzyme can be produced by culturing a strain belonging to Trachyderma genus such as Trachyderma K-2593 (FERM BP-387), etc. preferably at 20-25 deg.C and 4-7pH for 3-7 days under aeration and agitation. |
